2020年2月,来自浙江大学的颉孝贤等研究者在《情感障碍杂志》(Journal of Affective Disorders)上发表了研究,研究发现NMN(β-烟酰胺单核苷酸)可以改善小鼠的抑郁行为,具体表现为小鼠在游泳测试中获得更积极的结果,以及烟酰胺单核苷酸促进小鼠细胞产生能量。
厌食和体重减轻是常见的抑郁症状。该研究小组利用小鼠进行了相关实验,并发现抑郁小鼠的体重低于健康时的体重;而通过NMN注射,可以使体重恢复正常水平。研究人员通过向小鼠注射皮质酮(一种“压力荷尔蒙”)来诱发小鼠的抑郁行为。在注射了四个星期的皮质酮后(如下图蓝线所示),小鼠体重明显减轻,但将NMN添加到注射物中两周后(如红线所示),小鼠的体重可迅速恢复,几乎接近正常水平。

该研究还发现NMN可以让注射皮质酮的小鼠表现出更少的抑郁相关行为。科学家通过“强迫游泳测试”来模拟并且测量类似抑郁的行为。在这类测试中,小鼠被放在水中,为了浮在水面上,健康的小鼠将在水池中拼命游动,而抑郁、沮丧的小鼠会更快地选择放弃。补充NMN的抑郁小鼠则在水中变得更加积极,求生意愿更强。

研究在细胞层面也有所发现。三磷酸腺苷(ATP)是生物体内最直接的能量分子,负责储存和传递化学能,而过去的研究发现,抑郁会导致体内ATP的产生减少。这可能是因为细胞内负责制造能量的“能量工厂”线粒体发生了功能障碍。因此,提升线粒体的功能,可能是改善抑郁行为的有效思路。通过补充NMN等提高NAD+(烟酰胺腺嘌呤二核苷酸)水平,可以提升线粒体功能。
根据该项研究,NMN注射增强了大脑中线粒体的能量产生,并与抑郁行为的恢复有关。注射皮质酮的“抑郁小鼠”大脑中NAD+的水平降低了,但通过补充NMN,小鼠恢复了正常的NAD+的水平。
此外,该科学家团队还研究了NMN对调节细胞能量制造的蛋白质活性的影响。一种称为SIRT3的蛋白质对调节线粒体健康起着重要作用,SIRT3只有在健康水平的NAD+条件下才能发挥功能。最近的多项研究也发现该蛋白质与抑郁行为有关。当小鼠抑郁时,SIRT3蛋白的活性降低,而通过补充NMN,SIRT3活性得到改善,小鼠抑郁行为症状减轻,例如在“强迫游泳测试”中表现更好。
“我们的发现表明,补充NMN可以有效增强SIRT3蛋白活性,改善海马体(脑中负责短期记忆、长期记忆,以及空间定位的重要部分)和肝脏中的线粒体能量代谢,从而减轻皮质酮诱发的小鼠抑郁行为。”研究人员说。
科学家们同时也指出,该研究还存在着局限性,尤其是实验样本量较小的问题。研究人员也呼吁进行更大样本量的长期研究,以进一步研究NMN对抑郁行为的影响机制。
